چکیده

Ryruvate-phosphate dikmase formation was induced on transfer of glucose-grown cells of Acefobacfer xylinum to succinateor pyruvate-containing media. Enzyme formation did not occur if such media were supplemented with glucose or in the presence of chloramphenicol. Fructose and glycerol were less effective in repressing enzyme formation. When glucose was added to cultures of cells growing on succinate, the growth of the organism was unimpaired, but was accompanied by virtual cessation of dikinase synthesis. The changes in the dikinase activity of the cells were accompanied by parallel changes in the ability of cells to convert pyruvate into cellulose. It is concluded that the physiological function of pyruvate-phosphate dikinase in A. xylinum is gluconeogenic, and that gluconeogenesis in this organism is subject to control exerted over the synthesis of the dikinase.

Factors afecting the activity of pyruvate kinase of Acetobacter xylinum.

1. Extracts of Acetobacter xylinum were found to contain the glycolytic enzymes involved in the conversion of triose phosphate into pyruvate. Pyruvate kinase had the lowest relative activity. Phosphofructokinase activity was not detected in the extracts. 2. Only slight differences in the activity of pyruvate kinase were observed between cells grown on glucose and those grown on intermediates of...

Regulation of hexose phosphate metabolism in Acetobacter xylinum.

The metabolism of glucose and fructose was studied in resting succinate-grown cells of Acetobacter xylinum. From fructose only cellulose and CO(2) were formed by the cells, whereas from glucose, gluconate was formed much more rapidly than these two products. The molar ratio of sugar converted into cellulose to sugar converted into CO(2) was significantly greater than unity for both hexoses. The...

The dissimilation of glucose and gluconate by Acetobacter xylinum. 1. The origin and the fate of triose phosphate.

As shown by White & Wang (1964), A. xylinum, ATCC 10821, oxidizes glucose and gluconate to pyruvate and carbon dioxide by two pathways, the Entner-Doudoroff system and the pentose phosphate cycle. A fraction of the triose phosphate formed from these reactions recombines via aldolase to yield hexose phosphate which is metabolized further via the pentose phosphate cycle and the Entner-Doudoroff p...

Structural and evolutionary characteristics of pyruvate phosphate dikinase in Giardia lamblia and other amitochondriate protozoa.

BACKGROUND Pyruvate phosphate dikinase (PPDK) reversibly catalyzes the interconversion of phosphoenolpyruvate (PEP) and pyruvic acid, leading to catabolism and adenosine triphosphate (ATP) synthesis or gluconeogenesis and ATP consumption. Molecular modeling of PPDKs from divergent organisms demonstrates that the orientation of the phosphorylatable histidine residue within the central domain of ...
